General timeline for pronuclear injections
The following time line represents a general time frame for the production of a transgenic mouse using an FVB animal or other strain with good reproductive performance. Time frames are minimum and are intended to provide a general frame of reference in project planning. Some projects will require additional time. However, the core will keep you advised of the progress of your project.
Upon receipt of DNA sample and all associated paperwork, the core will initiate work on the specific project as follows:

- 1 2 weeks: 7 10 days are needed to order, receive and acclimate donor females.
- 2. 1 2 weeks: A minimum of 2 injection days are scheduled for each project. Additional injection days may be needed based upon number of zygotes recovered. For inbred strains other than FVB, we anticipate a minimum of 4 injection days. Typically two days a week are assigned for pronuclear injections.
- 3 5 weeks: Three weeks after embryo transfer, pups will be born. If injections were performed over 2 weeks, please allow 4 weeks from first injection for update on birth of pups. Pups are not handled during the first week after they are born to minimize stress to the mom.
- 3 4 weeks: Pups are weaned at approximately 3 weeks of age. At this time, animals are ear tagged, sexed, and tail samples are acquired. Tail samples are stored frozen until scheduled for analysis.
- 1 2 weeks. Once all tail samples are available, samples are processed for DNA and PCR is performed. Typically we will verify an internal gene and screen for gene of interest. Confirmation of results via repeat PCR is standard.
- 1 week: Once offspring are confirmed positive, the associated dam is submitted for health evaluation. When health report is received and approved, the Office of Animal Resources (OAR) will be contacted to facilitate transfer of mice to Investigator.

The overall time frame for the production of a transgenic pup ranges from a minimum of 10- 13 weeks. Additional time should be anticipated when inbred strains other than the FVB are used as these strains often produce low number of zygotes and will required multiple days of injection with low numbers to. Although this approach may be initially slower, if successful they eliminate the need for extensive back crossing.
