Transgenic Animal Core (TAC)

As of March 1, 2005, the TAC has worked with 13 constructs and generated over 100 transgenic pups.

SERVICE REQUEST PROCEDURES

Establishing a Transgenic Project or Knockout Project
1. Contact either Dr. Critser or Dr. Karasseva to discuss your project needs. (click here for contact info)

2. Complete a Request for Transgenic Mouse Production Form or Request for Knockout Mouse Form. This PDF form can be downloaded and filled out electronically. Then it must be printed, signed and sent to the Core by FAX or mail. Core FAX number is 573-884-7521. (click here to download PDF forms)

Investigators within the University of Missouri are advised that transgenic animals are produced under a sublicense agreement with Xenogen Biosciences. The transgenic animals are made for your research use only. Please read the terms of the sublicense agreement to ensure that you are in compliance with the terms of this agreement. Completion of a "Request for Transgenic Mouse" form indicates that you have read and understand this agreement. Transfer of transgenic mice to another investigator (on or off campus) requires the completion of a Material Transfer Agreement (MTA) document. (Click here to download MTA form). Please send the completed request form to the TAC by campus mail or FAX to 573-884-7521.

Investigators outside of the University of Missouri must complete a MTA document before any animal work can be initiated. (click here to download MTA form). In addition, please complete a Request for Transgenic Mouse form. Be sure to include a Purchase Order number and an ACUC approval number.

3. Prepare and provide DNA of appropriate quantity and quality, with map and photo of minigel.

4.
Provide PCR primers and protocol to TAC for transgenic work.

5. For transgenic work, the TAC will advise you as follows:
• Core will complete final DNA purification and advise if any concerns
were identified
• Core will confirm initial injection days scheduled for your project
• Core will confirm delivery of pups and advise of time frame for
pup screening.
• Core will report initial screening of tail DNA by PCR.
• Any adverse events during this process will be noted and reported.


For knockout projects, the TAC will advise you as follows:
• Core will perform electroporation, positive-negative selection and expansion, and advise when ES cell clones are ready for DNA purification and screening.
• After appropriate clones are identified, core will schedule expansion of clones, perform blastocyst injections and embryo transfers.
• Core will confirm delivery of pups and evaluate for coat color chimerism.
•All chimeric animals will be held until health monitoring is complete, at which time the chimeric animals will be transferred to the investigator.
