Transgenic Animal Core (TAC)


DNA purification guidelines

The Transgenic Animal Core asks the investigator to provide at least .1 mg of plasmid DNA purified with QIAGEN plasmid maxi kit (cat#12162 for 10 samples or cat#12163 for 25 samples). You can purchase the kit from the DNA Core here at MU campus (link to DNA core) or directly from QIAGEN. Since the purity of DNA sample is critical for successful generation of transgenic mice, please, DO NOT SUBSTITUTE the QIAGEN kit for another one or any other lab protocol.

If for some reason the QIAGEN kit cannot be used in your laboratory, please contact Dr. Karasseva for further discussion (KarassevaN@missouri.edu). The Transgenic Animal Core will perform additional endotoxin removal purification of your plasmid DNA as well as restriction digest of your construct to release the transgene.

The DNA fragment carrying the transgene will be cleaned up by the TAC to the quality compatible with microinjection. The Transgenic Animal Core requests the investigator to provide a map indicating the restriction sites for the transgene and the size of the insert.
